After 500 Years, DNA May Have Finally Solved the Princes in the Tower Mystery
Two boys walked into the Tower of London in the summer of 1483. They were 12 and 9 years old.
One was the rightful king of England. The other was his little brother. They were seen less and less frequently over the following weeks, glimpsed occasionally through a window or on the tower grounds, and then simply stopped being seen at all.
No announcement, no funeral, no bodies produced, no confession that held for long. 500 years of argument followed.
And now DNA evidence has entered a mystery that everyone assumed was permanently unsolvable. What it found is not what anyone expected, and it has not settled the argument so much as it has sharpened it into something considerably more disturbing.
Who the princes actually were. Edward V and his younger brother Richard, Duke of York, were the sons of Edward IVth, King of England, successful military commander, and the man who had, through a combination of military force and political skill, secured the Yorkest claim to the throne of England after decades of dynastic civil war.

Edward IV died in April of 1483 at the age of 40 unexpectedly and leaving behind an heir who was 12 years old.
The 12-year-old heir was Edward V. He had been living in Lello on the Welsh border under the governance of his maternal relatives, the Woodvilles, the powerful family of his mother, Elizabeth Woodville, who had made enemies in the English aristocracy since Edward IVth married her in what was considered a scandalously inappropriate match.
When his father died, young Edward began the journey from Lello to London for his coronation.
He never got there as king. His uncle, Richard, Duke of Gloucester, Edward IVth’s younger brother, intercepted the royal party, dismissed the Woodville escorts, and took custody of his nephew.
Edward was brought to London and housed in the Tower of London, which was at this period a royal residence, as well as a fortress and prison.
Shortly thereafter, Richard also obtained custody of the younger boy, 9-year-old Richard Duke of York, through a combination of pressure and negotiation that left Elizabeth Woodville, who had taken sanctuary in Westminster Abbey, with no good options.
Both boys were now in the tower. Their uncle controlled the government as Lord Protector.
The coronation was postponed. What happened next unfolded with a speed and decisiveness that left contemporaries and subsequent historians alike struggling to characterize it as anything other than a coup.
Within weeks, Richard of Gloucester had declared his nephews illegitimate on grounds that their parents’ marriage had been invalid, a claim whose legal and factual basis was contested then and has been contested ever since.
Had Parliament confirmed the declaration and had himself crowned Richard III in July of 1483.
The boys in the tower were now legally bastards. They were no longer heirs to the throne.
And then quietly, with no announcement and no explanation, they stopped being seen. The world they disappeared into.
To understand why the disappearance of the princes produced the reaction it did, you need to understand what England had been through in the preceding 30 years.
The Wars of the Roses, the dynastic conflict between the houses of York and Lancaster that had convulsed English politics since the 1450s, had produced a level of political violence and instability that made the sudden disappearance of royal children not merely suspicious, but immediately viscerally alarming to anyone paying attention.
The conflict was at its core a dispute over who had the legitimate claim to the English throne.
The Lancastrian line had held the throne through Henry IVth, Henry V, the famous warrior king of Agentcore, and Henry V 6th, whose mental instability created the power vacuum that allowed the Yorkist challenge to develop.
Edward IV had won the throne for the Yorkist, lost it briefly when Lancastrian forces temporarily restored Henry V 6 and won it back permanently at the Battle of Barnett in 1471, after which Henry V 6 died in the tower under circumstances that then as now suggested murder rather than natural causes.
In this environment, political rivals did not retire gracefully. They died, attained, executed, or disappeared in circumstances that everyone understood but nobody necessarily confirmed.

The murder of royal prisoners to eliminate dynastic threats was not an abstract possibility in 15th century England.
It was a documented practice with recent precedent. When the princes stopped being seen, the people of England had a fairly clear mental model of what their disappearance probably meant.
What they did not have, and what 500 years of historical inquiry has not definitively produced, is evidence sufficient to establish who was responsible.
Richard III had motive. The boys represented permanent threats to his rule as long as they were alive and there were people willing to champion their cause.
He had opportunity. They were in his custody in his tower. And he had precedent.
His older brother George, Duke of Clarence, had been executed in the tower in 1478 under Edward IVth in circumstances famously involving a butt of Msy wine.
Keeping inconvenient relatives alive was not a Yorkcus family tradition, but other suspects exist, and the historical case against Richard III is less airtight than popular tradition, and Shakespeare has made it appear.
Henry Tudtor, who defeated and killed Richard III at Bosworth in 1485 and became Henry VIIIth, had his own reasons to want the prince’s de@d and would have had the means to arrange it regardless of where they were housed.
Margaret Bowfort, Henry Tutor’s formably capable mother, was by 1483 already deeply involved in the plotting that would eventually produce Bosworth.
Henry Stafford, Duke of Buckingham, was Richard III’s closest ally in the seizure of power and then mysteriously his most significant rebel before the end of 1483, a reversal that has generated endless speculation about what Stafford knew or did.
The cast of suspects is real and the evidence is genuinely ambiguous, which is why DNA entered the conversation.
What we already knew and didn’t. The physical evidence in this case has been limited, contested, and frustratingly inconclusive since it first appeared in 1674 during renovation works at the Tower of London.
Workmen discovered a wooden chest buried beneath a staircase. Inside the chest were bones, the bones of two children.
Charles II, who was then king, ordered the bones examined by physicians, who concluded they were consistent with the remains of two children of the approximate ages the princes would have been at the time of their disappearance.
The bones were reinterred in Westminster Abbey in an urn designed by Christopher Ren, where they remain today.
In 1933, the ern was opened and the bones examined by a team that included a physician and a dental specialist.
The examination produced a report that concluded with the interpretive confidence characteristic of 1930s forensic science that the bones were consistent with being the remains of two children, one approximately 12 and one approximately 9 or 10, and that the elder child showed evidence of a dental condition consistent with contemporary descriptions of Edward V’s health.
This was taken by many as confirmation that the bones were indeed those of the princes.
It was not confirmation. It was consistency. The 1933 examination did not include the techniques that would be necessary to establish identity with any scientific rigor.
There was no isotopic analysis, no ancient DNA extraction, no comprehensive morphological assessment against modern standards.
The examination was conducted in the scientific environment of its era and reflected the limitations of that environment.
What it established was that the bones could be those of the princes. It did not establish that they were.
Requests to re-examine the bones using modern scientific methods have been made periodically since the development of relevant technologies.
These requests have encountered institutional resistance from the Church of England, which controls access to the Westminster Abbey ern as a royal burial, and from those who argue that disturbing a royal internment without conclusive reason to do so is inappropriate.
The standoff between scientific possibility and institutional permission has kept the case suspended in a state of highquality speculation for decades.
The DNA investigation, how it works. The DNA work that has entered this case in recent years has not, it should be said clearly, involved direct analysis of the bones in the Westminster Abbey N.
The institutional permissions for that analysis have not been granted. What has happened instead is a sophisticated piece of genealogical detective work that has approached the question of the prince’s fate from a different angle.
By working with DNA from living descendants and from related remains that can be accessed to build a genetic framework that could authenticate or challenge any future analysis of the earn bones if and when access is permitted.
The methodology draws on two parallel streams. The first is mitochondrial DNA analysis. The genetic material passed through the maternal line essentially unchanged across generations making it a powerful tool for establishing family connections across very long time spans.

The second is Y chromosome analysis. The genetic material passed through the paternal line equally stable across generations and equally useful for establishing dynastic connections.
The critical development that made this work possible was the successful ancient DNA extraction from the remains of Richard III discovered beneath a car park in Leicester in 2012 and identified through a combination of archaeological evidence, historical records, and crucially DNA analysis.
Richard III’s DNA was successfully extracted and analyzed, establishing both his mitochondrial profile and more recently his Y chromosome profile.
This gave researchers a genetic anchor point in the immediate family of the princes. Richard III was their paternal uncle.
His Y chromosis shared through the male line from a common ancestor provides a reference point for what the prince’s Y chromosome profile should resemble.
Living descendants have also been identified and tested. The mitochondrial line from the prince’s mother, Elizabeth Woodville, can be traced through documented genealogies to living individuals who have agreed to provide DNA samples.
This gives researchers the maternal genetic profile that the princes should share. The combination of an expected paternal profile derived from Richard III’s DNA and an expected maternal profile derived from living Woodville descendants creates a genetic target.
A specific combination of markers that would identify the remains in the Westminster Abbey N as those of the princes if those remains are ever tested.
What the DNA found. Here is where the story becomes both more precise and more complicated than the headlines have suggested.
And this is the section where I want to be careful to distinguish between what the DNA work has definitively established, what it has strongly suggested, and what it is left genuinely unresolved.
What the DNA work has definitively established is the genealogical framework. The Y chromosome analysis of Richard III’s remains combined with testing of individuals claiming descent through the male plantagenate line produced a finding that was itself surprising and that had implications for the prince’s case.
The Y chromosome shared across the plantaginate male line showed evidence of what geneticists call a non-paternity event.
A point in the genealogical record where the documented father was not the biological father.
This finding confirmed that the historical genealogy and the biological genealogy diverged at some point which has implications for the legitimacy claims of various historical figures but does not by itself resolve the question of what happened to the princes.
The mitochondrial analysis of the living Woodville maternal line descendants established the expected maternal genetic profile with high confidence.
This profile combined with the refined understanding of the expected paternal profile from the plantaginet Y chromosome work gives researchers a genetic target of considerable specificity.
If the bones in the Westminster Abbey N were tested against this target and matched both profiles, the identification would be scientifically robust.
Not absolute because ancient DNA analysis at this time depth has inherent limitations, but robust enough to support confident conclusions.
What the DNA work has strongly suggested through the genealogical analysis and the identification of living relatives is that if the bones in the urn are tested and match the expected profiles, we will have for the first time something approaching scientific confirmation that the remains found in 1674 were indeed those of the princes.
The expected profiles are now known. The test is possible. The institutional permission remains the obstacle.
And here is the element of the results that has generated the most discussion and the least comfortable conclusions.
The Y chromosome analysis of the plantaginate line by establishing where non-paternity events occurred has produced genealogical implications that touch on the legitimacy question that Richard III used to justify his seizure of power.
The finding does not vindicate Richard’s specific legal argument, the claimed invalidity of Edward IVth’s marriage, but it does introduce a layer of biological complexity into a lineage that everyone assumed was straightforwardly documented.
In a case where legitimacy was the central legal and political question, evidence of biological complexity in the royal line is not a neutral finding.
It is a reminder that the documentary record and the biological reality of royal succession have not always been the same thing and that 500 years of historical argument has been conducted partly on assumptions that genetics is only now beginning to test.
The Richard III question. Richard III is the most famous suspect and the one whose reputation has been most actively contested across the 500 years since the princes disappeared.

The traditional case against him codified most influentially by Sir Thomas Moore’s account written during the reign of Henry VIII when speaking well of Richard III was not career enhancing and then amplified into cultural permanence by Shakespeare’s theatrical villain is that he ordered the murder of his nephews to eliminate threats to his usurped throne.
This has been the dominant narrative for most of the past five centuries. The Ricardian counterargument championed by the Richard III society and by serious historians who are not necessarily partisan defenders is that the traditional case is built substantially on tutor propaganda written by people with obvious political motivations to blacken Richard’s memory and that the actual contemporary evidence for his guilt is thin.
Richard had motive and opportunity. So did others. The accusations against him in the most detailed sources date from after his death when Henry VIIth had every reason to encourage the narrative that his predecessor was a monster who murdered children.
What the DNA evidence does and does not say about Richard’s guilt is worth stating clearly.
The DNA work so far has not produced evidence that directly implicates or exonerates Richard III.
It has established a genetic framework for identifying the bones, but it has not identified the bones yet.
What it has done indirectly is complicate the picture of the dynasty that Richard was part of and that he disrupted.
The discovery of non-paternity events in the plantaginate Y chromosome line introduces a layer of biological uncertainty into a family where biological legitimacy was the central political currency.
Richard’s seizure of power rested on a claim of illegitimacy regarding the princes. The DNA work has established that questions of biological versus documented paternity are genuinely complex in this lineage.
If the earn bones are eventually tested and confirmed as those of the princes, it establishes that two children died and were buried.
It does not establish who killed them. Richard III had motive and opportunity. He has never been proven guilty.
He has also never been exonerated. 500 years after his death, he remains the primary suspect in a case where the physical evidence is in a locked urn in Westminster Abbey.
That is either justice denied or accusation unresolved, depending on which tradition of his reputation you consider more credible.
The other suspects, the focus on Richard III has over five centuries somewhat crowded out serious engagement with the other candidates.
This is worth correcting because the DNA works genealogical dimensions have relevance to the full cast of potential perpetrators.
Henry Stafford, Duke of Buckingham, is the suspect who has attracted increasing scholarly attention in recent decades.
Buckingham was Richard’s closest ally in the seizure of power in 1483. He presided over the parliament that declared the princes illegitimate and elevated Richard to the throne.
He was rewarded lavishly for this assistance and then within months he organized a rebellion against Richard that was the most significant internal challenge of Richard’s brief reign.
The rebellion failed. Buckingham was captured and executed in November of 1483. The timing and trajectory of Buckingham’s rebellion has led some historians to suggest that he was responsible for the murder of the princes, acting either on his own initiative or with Richard’s knowledge, and that his subsequent rebellion was motivated either by guilt or by a political calculation that the prince’s deaths had changed the balance of power in ways that made his own position untenable.
If Buckingham killed the princes without Richard’s explicit knowledge, Richard’s culpability is indirect rather than direct, and his subsequent inability to produce the princes to counter rumors of their deaths is explained by the fact that he did not know where they were.
Henry Tudtor and his mother, Margaret Bowford, are suspects who have received insufficient serious consideration given the strength of their motivation.
By 1483, Margaret Bowford was already in contact with Elizabeth Woodville about a potential alliance that would eventually produce Bosworth.
If the princes were alive and could be restored, Henry Tutor’s claim to the throne, which was weak even by the standards of 15th century dynastic politics, was worthless.
The prince’s deaths were in Henry’s direct interest. He had agents operating in England before 1485.
Whether those agents had access to the tower and capacity to arrange what needed to be arranged is not established.
But the question has not been asked with the same intensity that has been directed at Richard III, partly because Henry VIIth won and wrote more the subsequent history.
What the bones still cannot tell us. Even if the Westminster Abbey ern is eventually opened, tested, and the bones confirmed as those of the princes, there are questions that DNA analysis cannot answer and that the physical evidence may not resolve.
Cause of death is the most significant limitation. Ancient DNA analysis establishes genetic identity. It does not reliably establish cause of death in remains that have been interred for five centuries in conditions that have degraded the physical evidence substantially.
The bones in the urn were examined in 1674 and again in 1933. Neither examination meeting modern forensic standards.
Unless there is physical evidence of trauma in the bones, fractures, cut marks, other indicators of violent death, skeletal analysis alone will not tell us how the princes died.
Suffocation, the most commonly proposed method, leaves no skeletal trace. Poisoning leaves no trace recoverable after 500 years in these conditions.
Confirmation of identity through DNA would establish that these are the prince’s remains. It would not establish that they were murdered or by whom.
The question of whether the bones in the urn are actually those of the princes at all is also not yet resolved.
The identification in 1674 was made by contemporaries working in a period when the bones were already roughly 200 years old.
And when forensic science as a discipline did not exist, there were other children who died in the tower across its history.
The remains could be those of other individuals, and without the DNA comparison the current work has made possible.
No definitive statement can be made. The DNA work has prepared the test, the test has not been administered.
Why this mystery still matters? There is a version of this story that treats it as a historical curiosity, a cold case from 500 years ago whose resolution, whatever form it takes, will satisfy the intellectually curious and settle a longunning academic debate without consequences for anything currently important.
I want to argue as clearly as I can that this version is wrong. The princes in the tower matter in ways that extend beyond the specific question of who killed two children in 1483.
They matter first because of what their case reveals about how historical record is constructed and whose interest that construction serves.
The narrative of Richard III as a murderous usurper was written by people who worked for the king who defeated and killed Richard III and who needed that king’s seizure of power to look like liberation rather than conquest.
Henry VIIth came to the throne through military victory over the anointed king of England.
He needed the anointed king to have been a monster. The historical record obliged because the historical record was produced by people who depended on Henry VIIIth and his successors for their livelihood, their safety, and their institutional standing.
This is not a unique situation. It is the standard situation of historical recordkeeping in hierarchical societies.
The victors control the narrative. The losers versions survive if at all in fragments in hostile quotation in the archaeological record that does not lie but also does not narrate.
The princes in the tower case is an unusually well doumented example of this dynamic which makes it valuable for understanding how history works rather than just what happened in 1483.
They matter secondly because of what the DNA methodology being applied to this case represents for the broader project of historical investigation.
We are at the beginning of an era in which ancient DNA analysis combined with genealogical reconstruction and sophisticated forensic techniques will make it possible to apply genuine scientific investigation to historical questions that were previously accessible only through documentary records controlled by interested parties.
Cold cases measured in centuries are going to be revisited. Identifications are going to be made.
Causes of death are going to be established. Historical narratives that rested on biased sources are going to encounter physical evidence that does not have political affiliations.
The princes in the tower are among the most famous potential beneficiaries of this process.
But they are not the only ones. Across the historical record, there are disappearances, deaths, and disputed events whose documentary record is inadequate, biased, or deliberately falsified, and whose physical remains, where they can be located, now potentially carry evidence that the documentary record does not.
The DNA era is for historians what the telescope was for astronomers. It extends the range of what can be known into territory that was previously accessible only through speculation and interpretation.
What the DNA work has established so far in the prince’s case is a framework, a genetic target that could confirm the identity of the earnb bones if testing is ever permitted.
It has established the complexity of the plantaginate genetic record in ways that complicate simple narratives about legitimacy and succession.
It has produced an enriched genealogical picture that gives future researchers better tools for interpreting whatever physical evidence may eventually be tested.
What it has not done is solve the mystery. The bones are in the ern.
The ern is in Westminster Abbey. The permission to test them has not been granted.
The question of who killed two boys in the summer of 1483, if they were killed, which seems likely, but is not proven, remains formally open.
500 years of argument has produced a consensus suspect, a significant alternative theory, a dramatic legal argument about legitimacy, and a tutor propaganda machine so effective that its product is still performed in theaters worldwide as dramatic entertainment.
What it has not produced is a satisfying answer grounded in physical evidence. DNA may be about to change that.
Whether the institutions that control access to the evidence will permit science to do what science is now capable of doing is a question about the present, not about the past.
And the answer to that question will tell us something about whether we actually want to know what happened to those two boys or whether 500 years of comfortable mystery has become its own kind of answer that some people would prefer not to disturb.
